    • 摘要: 细胞凋亡是研究最为深入的细胞程序性死亡方式。但是近年发现,细胞中尚存在非凋亡性的程序性死亡机制。以往认为,细胞坏死是被动的细胞事件并且难以有效控制。新近发现的程序性坏死(necroptosis)作为一种死亡受体介导的半胱氨酸天冬氨酸蛋白酶(caspase)非依赖性细胞死亡模式为细胞死亡机制学说带来了重大突破。许多研究报道相继表明,细胞程序性坏死同样是受细胞内信号因子严密调控的主动过程。心肌细胞的坏死和凋亡是心脏疾病的重要病理途径。本文综述了心肌缺血与心衰过程中程序性坏死的作用及其与心肌细胞凋亡的关系,并探讨了针对心肌细胞程序性坏死的潜在治疗方向。

       

      Abstract: It is known that apoptosis is an actively mediated programmed cell death. However, in recent years, non-apoptotic programmed cell death has been recognized. Necrosis has traditionally been regarded as passive and unregulated and the newly discovered necroptosis is a death receptor mediated caspase-independent cell death. A large number of studies showed that necroptosis is an active process which is regulated by intracellular signal factors and myocardial cell necrosis and apoptosis are important pathological processes of heart diseases. This review discusses the role of necroptosis in myocardial ischemia and heart failure, its relationship with apoptotic pathways and new therapeutic opportunities the regulated necroptosis presents.
