    • 摘要: 目的: 探讨老年大鼠血管功能的特点及其能量代谢和线粒体功能变化。方法: 将16只大鼠分为两组(n=8),即老年大鼠组和成年大鼠组。禁食12 h后,测量大鼠全血血糖水平和血浆胰岛素的水平。取大鼠胸主动脉制作成2~3 mm的血管环,连接到离体血管灌流及记录系统记录血管的舒张变化情况。取血管灌流液测定NO的含量。用Western blot法测量腺苷酸活化蛋白激酶(AMPKα1)的蛋白的表达。采用782型氧气消耗仪测定胸主动脉细胞内线粒体的氧气消耗。结果: 禁食12 h后,老年大鼠组全血血糖水平高于成年大鼠组[(5.70±0.03) vs.(4.21±0.06)mmol/L,P<0.05]。老年大鼠组血浆胰岛素水平高于成年大鼠组[(23.6±0.2) vs.(14.4±0.2)μU/ml,P<0.05]。老年大鼠胸主动脉舒张率下降显著[(0.41±0.01) vs.(0.79±0.01)%,P<0.05]。老年大鼠组NO的含量显著低于成年大鼠组 [(9.07±0.15) vs.(45.33±0.38)μmol/L,P<0.05]。老年大鼠组AMPKα1的蛋白表达显著低于成年大鼠组(P<0.05);线粒体氧气消耗,两组差异明显[(1.33±0.33) vs.(2.42±0.03)μmol/min/g,P<0.05]。结论: 老年大鼠组存在全身性胰岛素抵抗;血管舒张功能下降程度显著;AMPKα1蛋白的表达及线粒体氧气消耗能力下降,是血管功能下降的机制之一。

       

      Abstract: AIM: To explore the characteristics of vascular function and energy metabolism and mitochondrial function change in aged rats. METHODS: Sixteen rats were divided into two groups (n=8), namely, aged rat group and adult rat group. After 12 h of fasting, glucose and plasma insulin levels were tested. Aortic vascular rings ~2-3 mm were produced and connected to the aortic perfusion and recording system to record the diastolic changes. Blood vessel perfusate was taken to measure NO content. Western blot was used to measure the expression of AMP-activated protein kinase (AMPKα1) using 782-type oxygen consumption apparatus to measure the oxygen consumption of mitochondria. RESULTS: After 12 h of fasting, blood glucose and plasma insulin levels, were higher in aged rat group [(5.70±0.03) vs.(4.21±0.06)mmol/L, P<0.05], [(23.6±0.2) vs.(14.4±0.2)μU/ml, P<0.05]. Diastolic function in aged group decreased significantly [(0.41±0.01) vs.(0.79±0.01)%, P<0.059). For NO content, there was a significant difference between the aged rat group and adult rat group [(9.07±0.15) vs.(45.33±0.38) μmol/L, P<0.05]. The expression of AMPKα1 in the aged rat group decreased significantly (P<0.05). For oxygen consumption in mitochondria there was a significant difference between the two groups [(1.33±0.33) vs.(2.42±0.03) μmol/min/g, P<0.05). CONCLUSION: For the aged rat group, there was a systemic insulin resistance. Vascular diastolic function decreased significantly. AMPKα1 expression and mitochondrial oxygen consumption capacity decreased markedly. It may be one of the mechanisms for decrease of vascular function.
