    Influence of obestatin on cardiac function and urine volume in chronic heart failure rats

    • 摘要: 目的 观察Obestatin腹腔注射对前壁心梗致慢性心衰(CHF)大鼠模型的心功能及其尿量的影响。方法 36只SD大鼠随机分为6组:对照组(n=6)及CHF模型组(n=30),后者包括:假手术组、精氨酸加压素(AVP)组、生长激素释放肽(Ghrelin)组、低剂量Obestatin组、高剂量Obestatin组(n均=6)。通过结扎心脏前降支建立CHF大鼠模型。16周后行心脏超声检查,并收集各组大鼠的24 h尿量,随后每天1次腹腔注射:对照组注射生理盐水,其余5组大鼠分别注射生理盐水、AVP、Ghrelin以及低剂量Obestatin〔50 μg/(kg·d)〕和高剂量Obestatin〔100 μg/(kg·d)〕,注射4周后复查心脏超声并收集各组大鼠当天24 h尿量。结果 腹腔注射4周后,与注射前相比,高剂量Obestatin组大鼠心脏超声左心室射血分数(LVEF)显著升高,尿量显著增多(均P<0.05);AVP组大鼠LVEF显著下降(P<0.01),尿量显著下降(P<0.05);Ghrelin组大鼠心脏超声LVEF值显著升高(P<0.01),尿量无显著变化;对照组、生理盐水组、低剂量Obestatin组大鼠LVEF、尿量无显著变化。结论 高剂量Obestatin长期腹腔注射能明显改善心梗后心衰大鼠心功能,增加尿量。

       

      Abstract: AIM The present investigation studied the effects of obestatin on changes in cardiac function and urine volume in post infarction chronic heart failure (CHF) rats. METHODS Thirty-six rats were randomly equally divided into 6 groups. Five CHF groups were established by ligating the left anterior descending artery (LAD), whereas rats in the sham group underwent chest operation without LAD ligation. After a 16-week normal diet, left ventricular ejection fraction (LVEF) was measured by echocardiography and a 24-hour urine was collected to measure urine volume in all sham and CHF rats. RESULTS The sham group was given intraperitoneal injection (IP) of saline, while the other five CHF groups were given IP of saline, AVP, ghrelin, obestatin 50 μg/(kg·d) and obestatin 100 μg/(kg·d), respectively. After the 4-week IP, LVEF was measured by echocardiography and a 24-hour urine was collected to measure urine volume in all sham and CHF rats. Compared with baseline, after the 4-week treatment, LVEF and urine volume increased significantly in the obestatin 100 μg/(kg·d) treatment group (P<0.05). LVEF decreased (P<0.01) and the urine volume also decreased significantly (P<0.05) in the AVP treatment group. LVEF increased significantly (P<0.01) but there were no significant changes in urine volume in the Ghrelin treatment group. There was no significant change in LVEF or urine volume in the sham group given saline or the CHF group given saline or the CHF group given obestatin (50 μg/kg·d). CONCLUSION In summary, high dose IP of obestatin, in a dose of 100 μg/(kg·d), improves cardiac function and urine volume in surgically prepared CHF rats.
