    • 摘要: 目的: 设计合成一种新的红景天苷(salidroside,SDS)类似物—酪醇半乳糖苷(tyrosol galactoside,TG),进一步探讨其对心脑的保护作用并与红景天苷的保护作用进行比较。方法: 实验分为两个部分:分别采用大鼠大脑中动脉闭塞(MCAO)模型和大鼠心肌缺血/再灌注(I/R)模型,通过神经功能评分(NBS)和TTC染色,评价两者(SDS和TG)的脑保护功能。通过左室收缩压(LVSP)、左室等容收缩/舒张期压力上升或下降最大速率(±dp/dtmax)、血清中肌酸激酶(CK)的活性和丙二醛(MDA)的水平评价两者的心脏保护功能。结果: TG和SDS均能显著改善大鼠局灶性脑I/R损伤NBS,减少脑梗死容积,但二者无统计学差异。与SDS相比较,TG组的LVSP和±dp/dtmax显著提高(P<0.05),血清中CK的活性和MDA的浓度也显著降低(P<0.01)。结论: TG和SDS均具有明显的心脑保护作用,但前者的心肌保护作用强于后者。

       

      Abstract: AIM: To design and synthesize a new analogue of salidroside-tyrosol galactoside, observe its protective effects on focal cerebral and cardiac ischemia/reperfusion (I/R) injuries and compare its protective effects with those of salidroside. METHODS: Using the model of middle cerebral artery occlusion (MCAO), we evaluated functions of cerebral protection by neurological behavior score (NBS) and TTC staining. In the model of myocardial I/R, left ventricular systolic pressure (LVSP) and maximal first derivative of developed pressure (±dP/dtmax) were monitored throughout the experiments. Serum creatine kinase (CK) and malondialdehyde (MDA) were detected at the end of reperfusion for evaluation of cardiac protection. RESULTS: Tyrosol galactoside and salidroside significantly improved NBS and reduced cerebral infarction volume with no statistical difference. In myocardial protection, tyrosol galactoside significantly increased LVSP and (±dP/dtmax) and decreased activities of CK. Level of MDAI and its protective effect was stronger than that of salidroside. CONCLUSION: Tyrosol galactoside and salidroside both have obvious protective effects on heart and brain. However, compared with salidroside, tyrosol galactoside has stronger effects on myocardial protection.
