    • 摘要: 目的:对比研究单独和联合应用左旋精氨酸 (L-Arg)和四氢生物喋呤(BH4)后,对幼兔离体心脏缺血/再灌注损伤(MIRI)的保护效果。方法: 将48只幼兔随机分为4组:Ⅰ组(对照组);Ⅱ组(BH4组);Ⅲ组(L-Arg组);Ⅳ组(BH4+L-Arg组);每组12只。建立Langendorff模型灌注其离体心脏,加入不同成分的K-H液灌流30 min后,检测缺血前、缺血后再灌注30 min、60 min、120 min时的冠脉流量(CF),心肌NO含量及NOS活性;线粒体丙二醛(MDA) 含量和超氧化物歧化酶(SOD)活性;检测心肌HSP70表达水平。结果: 与Ⅰ组相比,其他3组心肌组织NO含量明显增高,线粒体MDA含量降低,SOD活性升高,HSP70表达明显增强(P<0.05)。组间比较:Ⅱ组与Ⅲ组组间无明显差异;Ⅳ组与Ⅱ组与Ⅲ组相比,CF明显增高,NO含量明显增高,线粒体MDA含量降低,SOD活性升高,HSP70表达增强(P<0.05)。结论: 含BH4与L-Arg的灌注液能通过促进内源性NO的产生,保护心肌细胞线粒体功能,减轻未成熟心肌MIRI。

       

      Abstract: AIM: To study the effects of single or combined administration of tetrahydrobiopterin (BH4) and L-2-amino-5-guanidinovaleric acid (L-Arg) on Langendorff immature rabbit heart models. METHODS: Forty-eight immature rabbits were randomly divided into three test groups and one control group (n=12 per group). The isolated immature rabbit hearts were perfused retrogradely in the Langendorff model. The hearts were perfused with 37℃ Krebs-Henseleit buffer (KHB) for 30 min at 70 cm H2O (perfusion pressure). Coronary flow (CF) was recorded before ischemia and 30 min, 60 min and 120 min of reperfusion. The dynamic changes of myocardial NOS activity, NO content, mitochondrial content of malonaldehyde (MDA) and superoxide dismutase (SOD) were measured at different points. The expression level of myocardial HSP70 was also detected. RESULTS: Compared with that in the control group, the production of NO and SOD significantly increased in other groups, and the production of MDA was significantly degraded in other groups. Compared with that in group II and group III, there were no differences in the production of NO and SOD and MDA. Compared with that in group II and group III, the production of NO and SOD and CF significantly increased in group IV, and the production of MDA was significantly degraded. The expression level of myocardial HSP70 was significantly increased. CONCLUSION: Compared with the single administration, the combined administration of BH4 and L-Arg may have better cardioprotective effects by increasing NO and preserving the function of mitochondria.
