    跑台运动调节线粒体ATP释放改善高脂饮食大鼠的心肌损伤

    Treadmill exercise modulates mitochondrial ATP release to ameliorate myocardial injury in high-fat diet rats

      目的 探究跑台运动调节线粒体ATP释放改善高脂饮食大鼠的心肌损伤。
      方法 将大鼠随机分为Con组、HD组和AE组,每组10只。比较大鼠心功能(LVEF、LVFS、LVEDD、LVESD);比较大鼠生化指标(LDH、GPT活性及TG、TC含量);HE染色检测心肌组织病理学变化;检测大鼠ATP含量和线粒体膜电位变化;蛋白质印迹检测心肌组织中抵抗素和脂联素蛋白表达。
      结果 与Con组相比,HD组大鼠心功能指标LVEF和LVFS明显降低,LVEDD和LVESD明显升高(P<0.05);与HD组相比,AE组大鼠心功能指标LVEF和LVFS明显升高,LVEDD和LVESD明显降低(P<0.05)。与Con组相比,HD组大鼠血清中LDH、GPT活性及TG、TC含量明显升高(P<0.05);与HD组相比,AE组大鼠血清中LDH、GPT活性及TG、TC含量均明显降低(P<0.05)。与Con组相比,HD组大鼠心肌组织中ATP含量、心肌正常细胞数和线粒体膜电位未降低细胞的百分比均明显降低(P<0.05);与HD组相比,AE组大鼠心肌组织中ATP含量、心肌正常细胞数和线粒体膜电位未降低细胞的百分比均明显升高(P<0.05)。与Con组相比,HD组大鼠心肌组织中抵抗素蛋白表达明显增加,脂联素蛋白表达明显降低(P<0.05);与HD组相比,AE组大鼠心肌组织中抵抗素蛋白表达明显降低,脂联素蛋白表达明显升高(P<0.05)。
      结论 跑台运动可改善高脂饮食大鼠心肌损伤,其可能是通过增加线粒体ATP的释放而发挥保护作用。

       

      Abstract:
      AIM To explore the improvement of myocardial injury in rats with high-fat diet through regulating mitochondrial ATP release by running.
      METHODS Rats were randomly divided into Con group, HD group and AE group, 10 rats in each group. Rat cardiac functions (LVEF, LVFS, LVEDD, LVESD) and rat biochemical indexes (LDH, GPT activity and TG, TC content) were compared. Myocardial histopathological changes were detected by HE staining, rat ATP content and mitochondrial membrane potential changes were detected and resitin and adiponectin expressions in myocardial tissue were detected by Western blot.
      RESULTS Compared with those in Con group, LVEF and LVFS of the rats in HD group were significantly decreased, while LVEDD and LVESD were significantly increased (P<0.05). Compared with those in HD group, LVEF and LVFS of the rats in AE group were significantly increased while LVEDD and LVESD were significantly decreased (P<0.05). Compared with those in Con group, the activities of LDH and GPT and the contents of TG and TC in the serum of the rats in HD group were significantly increased (P<0.05). Compared with those in Con group, the content of ATP, the number of normal myocardial cells and the percentage of cells with unreduced mitochondrial membrane potential in the myocardial tissue in HD group were significantly decreased (P<0.05). The content of ATP, the number of normal myocardial cells and the percentage of cells with no decrease in mitochondrial membrane potential were significantly increased (P<0.05). Compared with those in Con group, the expression of resistin in the myocardial tissue in HD group was significantly increased and the expression of adiponectin was significantly decreased (P<0.05). The protein expression was significantly increased (P<0.05).
      CONCLUSION Running can improve myocardial injury in rats with high-fat diet, which may play a protective role by increasing the release of mitochondrial ATP.
