    • 摘要: 线粒体-内质网接触点(mitochondria-endoplasmic reticulum contacts, MERCs)是线粒体与内质网之间通过多种连接蛋白形成的物理结构。MERCs在多种生理过程中发挥重要功能,如Ca2+与脂质转运、线粒体ATP合成、线粒体融合与分裂以及线粒体自噬等。在病理条件下,MERCs部位线粒体外膜的电压依赖性阴离子通道(VDAC)开放,使线粒体膜间腔中的凋亡诱导因子如细胞色素C释放,导致细胞凋亡;或者线粒体通透性孔(MPTP)开放,使ROS、心磷脂与mtDNA等外溢出线粒体,在MERCs部位激活NLRP3炎性小体装配,从而作为炎症的起始点,诱发低强度且持续的炎性反应,在慢性心血管疾病的形成中发挥重要作用。因此,MERCs可作为心血管疾病治疗的重要靶点。本文介绍了MERCs的结构特征与功能,重点讨论了MERCs作为NLRP3炎性小体装配位点在多种心血管疾病中的作用。

       

      Abstract: Mitochondria-endoplasmic reticulum contacts (MERCs) are physical structures formed between mitochondria and endoplasmic reticulum through a variety of junctional proteins. MERCs play an important role in a variety of physiological processes, such as Ca2+ and lipid transport, mitochondrial ATP synthesis, mitochondrial fusion and fission, and mitophagy. Under pathological conditions, the voltage-dependent anion channel (VDAC) of the mitochondrial outer membrane at MERCs is open, which releases the apoptosis-inducing factors such as cytochrome C in the mitochondrial intercellular space, leading to cell apoptosis. Or the mitochondrial permeability transition pore (MPTP) is open, allowing ROS, cardiolipin and mtDNA to overflow the mitochondria and activating NLRP3 inflammasome assembly at the site of MERCs, thus inducing low-intensity and sustained inflammatory response as the starting point of inflammation, which plays an important role in the formation of chronic cardiovascular disease. Therefore, MERCs can be used as critical targets for the treatment of chronic cardiovascular diseases. This review introduces the structural characteristics and functions of MERCs and focuses on the role of MERCs as NLRP3 inflammasome assembly sites in a variety of cardiovascular diseases.
