    Research progress in mechanism of ferroptosis and its role in atherosclerosis

    • 摘要: 铁死亡是近年来发现的一种新型细胞程序性死亡方式,已被证实与心肌病、心力衰竭、动脉粥样硬化、缺血/再灌注损伤等多种心血管疾病相关。铁死亡的典型特征包括细胞内氧化还原活性铁增加、脂质过氧化及活性氧清除障碍。铁死亡可导致内皮细胞功能障碍,调控巨噬细胞死亡与表型极化,参与平滑肌细胞表型转换,促进动脉粥样硬化进展与斑块易损性增加。探究铁死亡在动脉粥样硬化中的作用,将为冠心病疾病防治与预后改善提供新的诊疗思路与药物研发靶点。本文将就铁死亡的发生与调控机制,及其在动脉粥样硬化中的最新研究进展进行综述。

       

      Abstract: Ferroptosis is a new type of programmed cell death discovered in recent years, which has been proven to be associated with cardiomyopathy, heart failure, atherosclerosis, ischemia/reperfusion injury and other cardiovascular diseases. The typical features of ferroptosis include excessive intracellular redox-active iron, lipid peroxidation and impaired clearance of reactive oxygen species. Ferroptosis could lead to endothelial dysfunction, regulate macrophage death and phenotypic polarization, impact the phenotypic switch of smooth muscle cells and promote atherosclerotic progression and increased plaque vulnerability. Exploring the role of ferroptosis in atherosclerosis will provide new diagnostic and treatment strategies and drug design targets for clinical management and prognosis improvement of coronary heart diseases. In this review, the advanced research progress in the mechanism of ferroptosis and its role in atherosclerosis is summarized.
