自制小动物定量创伤仪对大鼠心肌组织损伤的观察

    Observation of myocardial tissue injuries caused by a self-made, small animal quantitative trauma device

    • 摘要: 目的:观察使用自制小动物定量创伤仪,造成的大鼠全身机械性创伤。方法: 首先设计并组装制造实验用小动物定量全身性机械性创伤仪。将40 只大鼠分为对照组和创伤组,每组20只(n=20),以40 r/min的转速旋转200圈,分别给予创伤组动物定量创伤,观察心肌组织中心肌细胞的凋亡指数、Caspase-3、iNOS、NO、NADPH氧化酶以及超氧阴离子(·O-2)的表达量,对照组未做任何处理。结果: 创伤组心肌细胞的凋亡指数、Caspase-3以及iNOS的表达、NO的产生、NADPH氧化酶的表达及·O-2的生成,均明显高于对照组(P<0.01)。结论: 使用自制小动物创伤仪,定量给予大鼠全身性机械性创伤,可造成大鼠心肌细胞发生明显的氧化应激反应和凋亡。

       

      Abstract: AIM:To observe myocardial tissue injury caused by a self-made, small animal quantitative trauma device. METHODS: Forty rats were randomly divided equally into two groups: control group (n=20) and trauma group (n=20). Rats of the trauma group were anesthetized with pentobarbital sodium (40 mg/kg), individually placed on a self-made small animal quantitative trauma device and were traumatized as the wheel rotated (200 revolutions (r) at a rate of 40 r/min). Measures were taken to observe the pathological changes of myocardial tissue. RESULTS: Compared with those in control group, the apoptotic index of the myocardial cells was elevated (P<0.01). Activity of caspase-3, expression of both iNOS and NADPH oxidase, and production of both NO and ·O-2 (superoxide anion) were also increased in the trauma group (all P<0.01). CONCLUSION: Mechanical trauma may induce significant oxidative stress and apoptosis in rat myocardial tissues by using a self-made, small animal quantitative trauma device.
