铜稳态与心血管疾病:分子机制及病理生理作用

    Copper homeostasis and cardiovascular disease: molecular mechanisms and pathophysiological role

    • 摘要: 心血管疾病是全球人类死亡的主要原因,其发病率与病死率逐年上升,急需有效的干预与管理措施改善患者预后。尽管先前研究已探索心血管疾病的发病机制,但其具体的分子机制仍未充分阐明。铜离子是细胞内最重要的信号分子之一,其动态平衡参与了人体内氧化应激、能量代谢及信号传递等多种生物学过程,而铜缺乏或铜过载均可导致不良的心血管事件的发生与发展。最近的研究表明,铜死亡是不同于细胞凋亡、坏死性凋亡、焦亡、自噬与铁死亡的一种新型程序性死亡方式,在心脏重构、动脉粥样硬化、冠心病、心肌病、心律失常、心力衰竭等心血管系统的病理生理过程及疾病的进展中扮演重要作用。本文总结了铜稳态和铜死亡在心血管疾病中的分子机制及病理生理作用,有望为心血管疾病的诊断、治疗与预后提供新型靶点或策略。

       

      Abstract: Cardiovascular disease is the main cause of human death worldwide and its incidence rate and mortality are increasing year by year. Effective intervention and management measures are urgently needed to improve the prognosis of patients. Although previous studies have explored the pathogenesis of cardiovascular disease, its specific molecular mechanisms have not been fully elucidated. Copper ions are one of the most important signaling molecules in cells and their dynamic balance is involved in various biological processes such as oxidative stress, energy metabolism and signal transmission in the human body. Copper deficiency or overload can lead to the occurrence and development of adverse cardiovascular events. Recent studies have shown that cuproptosis is a new type of programmed cell death that is different from cell apoptosis, necroptosis, pyroptosis, autophagy and ferroptosis, and it is involved and plays an important role in cardiac remodeling, atherosclerosis, coronary heart disease, cardiomyopathy, arrhythmia, heart failure and other cardiovascular pathophysiological processes and disease development. This article summarizes the molecular mechanisms and pathophysiological roles of copper homeostasis and cuproptosis in cardiovascular diseases, which is expected to provide new targets or strategies for the diagnosis, treatment and prognosis of cardiovascular diseases.
