Effects of isopropyl 3-(3,4-dihydroxyphenyl)-2-hydroxypropanoate at different concentrations on left ventricular functions of chronic hypoxic rats

    • AIM: To observe the effects of isopropyl 3-(3,4-dihydroxyphenyl)-2-hydroxylpropanoate at different concentrations on the left ventricular function of chronic hypoxic rats. METHODS: Chronic hypoxic rats were equally divided into four groups: large-dose isopropyl 3-(3,4-dihydroxyphenyl)-2-hydroxylpropanoate group, small-dose isopropyl 3-(3,4-dihydroxyphenyl)-2-hydroxylpropanoate group, positive control group of verapamil and normal saline group. Homodynamic and heart function parameters were measured by intubating from left common carotid artery. RESULTS: Compared with those in normal group, SBP, DBP, MBP, LVSP, left ventricular +dp/dtmax and -dp/dtmax markedly increased (P<0.05 or 0.01), and t-dp/dtmax markedly decreased (P<0.05) in model group, HE staining revealed disordered arrange of myocardial cell, fracture and dissolution of muscle fibers and nuclear fission in model group. Compared with those in the model group, DBP, MBP, HR, LVEDP and -dp/dtmax markedly decreased (P<0.05 or 0.01), and t-dp/dtmax markedly increased (P<0.05) in the small-dose group, whereas SBP, DBP, MBP, HR, LVEDP, LVDP, +dp/dtmax and -dp/dtmax markedly decreased (P<0.05 or 0.01), and t-dp/dtmax markedly increased (P<0.01) in the large-dose group. HE staining revealed large amounts of glycogen in myocardial cells, no visible disordered arrangement of myocardial cell, fracture and dissolution of muscle fibers and nuclear fission in small- and large-dose group. Compared with those in the positive group, DBP, MBP, LVSP, LVDP and t-dp/dtmax markedly increased (P<0.05 or <0.01), and LVEDP markedly decreased (P<0.05) in the small-dose group, whereas DBP and t-dp/dtmax markedly increased (P<0.05 or <0.01), LVDP, LVEDP and +dp/dtmax markedly decreased (P<0.05 or <0.01) in the large-dose group. CONCLUSION: Isopropyl 3-(3,4-dihydroxyphenyl)-2-hydroxylpropanoate significantly inhibits high blood pressure, rapid heart rate and cardiac function excitement in chronic hypoxic rats. So it may significantly protect the cardiac function of chronic hypoxic rats.
