    Effect of advanced glycation end products on angiogenesis and tube formation of microvascular endothelial cells

    • AIM: To investigate the effects of the advanced glycation end products (AGEs) on the proliferation, migration and capillary tubule-like formation (TLS) in cardiac microvascular endothelial cells (CMECs). METHODS: CMECs were cultured with AGEs at different concentrations (100 mg/L, 200 mg/L, and 400 mg/L) for 48 h. MTT and transwell were used to detect the proliferation and migration and capillary tubule-like formation was examined to detect the angiogenesis. RESULTS: The A values of CMECs at the AGEs concentration of 100 mg/L, 200 mg/L and 400 mg/L after 48 h incubation were 0.1195±0.0049, 0.1422±0.0058 and 0.1783±0.0220, significantly higher than 0.0955±0.0161 (P<0.05, P<0.01). The migrations of CMECs (24.89±6.234, 32.89±6.990 and 55.56±10.27) were higher than 13.89±4.622 (P<0.05, P<0.01), and the tubule-like formation structures of CMECs were 3.261±0.7016, 4.737±1.129 and 6.687±1.308, higher than 2.089±0.6723 (P<0.05, P<0.01 vs. control group) in a concentration dependent manner. CONCLUSION: AGEs promote the proliferation, migration and tubule-like structure formation in a concentration-dependent manner.
