Qishen Yiqi Dropping Pill alleviates myocardial ischemia-reperfusion injury and its effect on the expression of AKT and p-AKT protein in myocardial tissue

  AIM  To investigate the effects of Qishen Yiqi Dropping Pills on alleviating acute myocardial ischemia-reperfusion injury in Bama pigs and its effects on the expression of protein kinase B(Akt) and phosphorylated protein kinase B(p-Akt).

  METHODS  Twenty healthy Guangxi Bama pigs were randomly divided into Qishen Yiqi Dropping Pill group (n = 10) and model group (n = 10). In addition to the normal feeding of Bama pigs in the two groups, Qishen Yiqi Dropping Pills group was given Qishen Yiqi Dropping Pills plus 100 ml pure water, while the model group was given 100 ml pure water by intragastric administration, 3 times a day for 2 months. The model of myocardial ischemia-reperfusion injury was established by ligating the anterior descending branch of the left coronary artery in Bama pigs. Serum CK, CK-MB and cTnT 2 were detected by ELISA method, the area of myocardial infarction was detected by triphenyltetrazolium chloride (TTC) staining and the expression levels of AKT and P-AKT in normal zone, marginal zone and infarcted area were detected by Western blot.

  RESULTS  ELISA showed that CK, CK-MB and cTnT values in Qishen Yiqi Qropping Pill group were all smaller than those in model group (P<0.05). TTC staining found that compared with those in model group, the area of myocardial infarction in Bama pigs in Qishen Yiqi Dropping Pills group was significantly reduced and the difference was statistically significant (P<0.05). Western blot detection results showed that no significant changes in the expression level of AKT protein were seen between model group and Qishen Yiqi Dropping Pills group, but the expression of P-AKT protein in Qishen Yiqi Dropping Pills group increased significantly and the difference was statistically significant (P<0.01).

  CONCLUSION  Qishen Yiqi Dropping Pills reduces the area of infarted myocardium and the content of markers of myocardial injury, and then alleviates the ischemia reperfusion injury of myocardial tissue, meanwhile, it activates the expression of p-Akt protein in cardiac tissue.
